摘要:
The highly esteemed Chinese herb, Dendrobium huoshanense, whose major metabolites are polysaccharides and alkaloids, is on the verge of extinction. The stone planting under the forest (SPUF) and greenhouse planting (GP) of D. huoshanense are two different cultivation methods of pharmaceutical Dendrobium with significantly differences in morphology, metabolites content and composition, and medication efficacy. Here, we conducted proteomics and phosphoproteomics analyses to reveal differences in molecular mechanisms between SPUF and GP. We identified 237 differentially expressed proteins (DEPs) between the two proteomes, and 291 modification sites belonging to 215 phosphoproteins with a phosphorylation level significantly changed (PLSC) were observed. GO, KEGG pathway, protein domain, and cluster analyses revealed that these DEPs were mainly localized in the chloroplast; involved in processes such as posttranslational modification, carbohydrate transport and metabolism, and secondary metabolite biosynthesis; and enriched in pathways mainly including linoleic acid metabolism, plant-pathogen interactions, and phenylpropanoid, cutin, suberin, and wax biosynthesis. PLSC phosphoproteins were mainly located in the chloroplast, and highly enriched in responses to different stresses and signal transduction mechanisms through protein kinase and phosphotransferase activities. Significant differences between SPUF and GP were observed by mapping the DEPs and phosphorylated proteins to photosynthesis and polysaccharide and alkaloid biosynthesis pathways. Phosphorylation characteristics and kinase categories in D. huoshanense were also clarified in this study. We analyzed different molecular mechanisms between SPUF and GP at proteomic and phosphoproteomic levels, providing valuable information for the development and utilization of D. huoshanense. 摘要:
沼液成分复杂,比较难处理。Fenton试剂法可以在反应中产生活性极强的羟基自由基,能无选择性地矿化有机污染物。采用Fenton试剂催化降解沼液,分别研究在不同pH值、不同催化剂投加量、不同H2O2投加量、不同Fe2+投加量及不同反应时间等实验条件下对沼液降解效果的影响,确定Fenton试剂催化降解沼液的最佳反应条件。实验结果标明:当pH值为3时,H2O2的浓度为15 mL/L,Fe2+的浓度为130 mg/L,反应时间控制在2 h,为催化降解沼液的最佳运行条件,沼液中的COD去除效果最好。

一株耐铀细菌的分离鉴定及其耐铀特性研究

摘要:
从湖南省衡阳市272厂铀尾矿库筛选分离出一株耐铀细菌Y-1,通过16S rRNA基因测序比对,初步鉴定该菌株为蜡样芽孢杆菌(Bacillus cereus).通过静态吸附实验,考察不同pH值、吸附时间、吸附剂投加量和初始铀浓度对菌株吸附U(VI)的影响.结果表明,该菌株具有良好的吸附效果,在吸附条件T=30℃,pH=5,t=200 min,C0(吸附剂)=0.3 g/L,C0(UO22+)=10 mg/L时,对U(VI)的去除率达到了89.5%.说明该菌株在铀污染的土壤生物修复方面具有潜在的应用前景.

摘要:
本实验旨在利用微生物发酵技术,以低廉的果皮为发酵原料,制备生物絮凝剂。采用暴露于空气中的红糖液作为菌种来源,通过平板划线对红糖液中菌株进行分离筛选,将纯化后的菌种添加至灭菌后的果皮上进行发酵处理,用发酵产物对高岭土悬液进行絮凝实验,菌株H1-3发酵形成的产物絮凝效果最佳。经形态观察及对比分析,该菌株为黑曲霉,通过单因素实验得出H1-3的最佳发酵条件为:发酵温度为35℃,发酵时间为72 h,菌种添加量为8%,摇床转速160 r/min。絮凝效果最佳的条件为絮凝剂投加量为1 mL。

随着铅锌矿区的不断开采,重金属随着各种介质进入土壤,使得某铅锌矿区土壤重金属含量超标,为了修复某铅锌矿区锌含量超标的土壤,本实验旨在探究香附子对锌的富集能力,以便利用植物修复的方法修复被污染的土壤.收集重金属土壤的香附子的根、茎、叶及土壤样品,利用浓硝酸-高氯酸处理植物样品,利用浓硝酸-氢氟酸-过氧化氢处理土壤样品,用原子吸收光谱分别测定植物及土壤样品待测液中锌的含量.实验结果表明,某铅锌矿区的土壤重金属含量超过国家标准,香附子的吸附系数,转运系数均大于1,因此,香附子对锌具有富集作用,植株各部分对锌的富集情况是:根>茎>叶.

摘要:
通过对蓝尾蝾螈皮肤进行转录组测序,首次报道了蓝尾蝾螈皮肤的转录组及其活性多肽成分.采用Illumina Hiseq4000共测定了6.65 Gb数据;组装和去冗余后共获得75 462个unigene,其N50、GC含量、平均长度、总长度分别为1 617 bp、45.64%、818 bp、61 801 999 bp.通过基因注释,共获得34 850个非冗余注释unigene(占unigene总数的46.18%),注释结果为两栖类物种的unigene数目约占注释总数的16%.根据unigene的COG和GO注释结果,统计了各自的功能分类.其中,对鉴定到的多种活性多肽进行了初步的生物信息学分析,为诠释蓝尾蝾螈皮肤生理学功能提供依据.

摘要:
研究异叶茴芹(Pimpinella diversifolia DC.)提取物对Ⅱ型糖尿病模型小鼠的血糖及相关因子的影响和作用。选取健康KM小鼠,建立Ⅱ型糖尿病模型,分高、中、低剂量组[160、80、40 mg(/kg·d)]以异叶茴芹提取物灌胃3周后,测定各组小鼠空腹血糖值、脏器指数以及血清中BUN、CHOL、GLU、TRIG水平。结果表明,异叶茴芹提取物可显著降低小鼠血糖值(P<0.05),其中,血糖降低程度为阳性组>高浓度组>中浓度组>低浓度组;在血清指数中,高中低3组与阳性组的差异不大,说明异叶茴芹提取物的作用与阳性药物一致。异叶茴芹提取物可改善Ⅱ型糖尿病小鼠血糖水平,提高机体抗氧化能力。

摘要:
Root hypodermis passage cells (PCs) lack suberin and lignin deposition, responsible for nutrient absorption and arbuscular mycorrhizal colonization, which are crucial for root resource acquisition. Nevertheless, their quantitative variability across diverse woody species and their relationships with root morphology and anatomy, as well as arbuscular mycorrhizal colonization, are still not well understood. Herein, the number and proportion of PCs in the root cross-section, root morphology, anatomy, and arbuscular mycorrhizal colonization rate were quantified across 10 temperate and 12 tropical woody species. The objectives of this study were to determine how PCs vary across contrasting environmental conditions and to explore their relationships with arbuscular mycorrhizal colonization rate and other root functional traits. The results showed that tropical species possessed 56% more PCs than temperate species; by contrast, they had similar proportions of PCs. In both biomes, the number of PCs had a tightly positive correlation with arbuscular mycorrhizal colonization rate (R2 = 0.35–0.87), root diameter (R2 = 0.84–0.93), and cortex thickness (R2 = 0.87–0.89), but the proportion of PCs was mostly independent of root morphological and anatomical traits. Our results suggest that variation in passage cells could well explain the tight linkage between arbuscular mycorrhizal colonization and root diameter across species and biomes, which provides insight into the collaboration gradient between plant roots and mycorrhizal fungi that dominates the root economics space. © 2022 by the authors. [目的]从加入农富康生物菌剂、农保有机肥发酵剂和强兴堆肥发酵剂发酵的猪粪好氧堆肥中,筛选出高效抑制臭气的菌株.[方法]以新鲜猪粪为原料接种发酵剂,通过选择培养基进行分离筛选,筛选出的菌株接种至新鲜猪粪检验其除臭效果.[结果]共筛选出3株可高效抑制臭气的菌株,分别标记为nf5、nb11和qx18.通过形态学和生理生化实验,鉴定该三种菌株:nf5为乳酸杆菌科乳酸杆菌科(Lactobacillaceae),nb11为曲霉科(Aspergillace-ae),qx18为酵母菌科(Saccharomycetaceae).[结论]将三种菌株1 ∶ 1复配优化,使用单因素法可以确定最佳接种条件为:猪粪含水率50%,最佳发酵温度32℃,接种量12%.单菌株中臭气抑制效果最佳的是nf5.

【目的】研究堆肥发酵剂对猪粪发酵的影响。【方法】以猪粪、锯木灰和堆肥发酵剂为原材料进行好氧堆肥,测定堆肥过程中的温度变化,分别以ITS1F_ITS2R和338F_806R作为真菌和细菌引物测定微生物丰富度和多样性。【结果】堆肥发酵剂能促进猪粪的堆肥发酵升温,增加微生物群落的丰富度和多样性并改变其优势种。【结论】对照组的真菌优势种有肉座菌科(Hypocreaceae)、曲霉科(Aspergillaceae),细菌优势种有梭菌科(Clostridiaceae)、乳酸杆菌科(Lactobacillaceae)和杆菌科(Bacillaceae),而试验组的真菌优势种有曲霉科和丝孢酵母科(Trichosporonaceae),细菌的优势种有梭菌科(Clostridiaceae)、消化链球菌科(Peptostreptococcaceae)、链球菌科(Streptococcaceae)。

摘要:
Platycodin D (PD) is a deglycosylated triterpene saponin with much higher pharmacological activity than glycosylated platycoside E (PE). Extensive studies in vitro showed that the transformation of platycoside E to platycodin D can be achieved using β-glucosidase extracted from several bacteria. However, whether similar enzymes in Platycodon grandiflorus could convert platycoside E to platycodin D, as well as the molecular mechanism underlying the deglycosylation process of platycodon E, remain unclear. Here, we identified a β-glucosidase in P. grandiflorus from our previous RNA-seq analysis, with a full-length cDNA of 1,488 bp encoding 495 amino acids. Bioinformatics and phylogenetic analyses showed that β-glucosidases in P. grandiflorus have high homology with other plant β-glucosidases. Subcellular localization showed that there is no subcellular preference for its encoding gene. β-glucosidase was successfully expressed as 6 × His-tagged fusion protein in Escherichia coli BL21 (DE3). Western blot analysis yielded a recombinant protein of approximately 68 kDa. In vitro enzymatic reactions determined that β-glucosidase was functional and could convert PE to PD. RT-qPCR analysis showed that the expression level of β-glucosidase was higher at night than during the day, with the highest expression level between 9:00 and 12:00 at night. Analysis of the promoter sequence showed many light-responsive cis-acting elements, suggesting that the light might regulate the gene. The results will contribute to the further study of the biosynthesis and metabolism regulation of triterpenoid saponins in P. grandiflorus. 摘要:
Phenolic compounds are bioactive metabolites in plants. The phenolic compounds in husks and seeds of areca nut (AN) were extracted via conventional liquid–solid extraction and ultrasonic-microwave synergistic extraction (UMSE), and abbreviated as CHP, CSP, UHP, and USP, respectively. The extraction conditions of UHP and USP were optimized (solid/solvent ratio of 1:40 and 1:50, the ethanol concentration of 50%, microwave power of 600 and 300 W, and extraction time of 360 and 540 s, respectively). The total phenolic content (TPC) yield, purity, antioxidant activity, and antibacterial activity of four samples were determined and compared. The USP has excellent TPC yield (270.92 mg GAE/g DW), purity (42.84%), antioxidant activities (the IC50 of DPPH, ABTS, and Superoxide anion were 55.40, 81.87, 138.65 µg/mL, respectively), and antibacterial activities (the MIC of Streptococcus mutans, Staphylococcus aureus, Escherichia coli, and Pseudomonas aeruginosa were 2.50, 5.00, 15.00, and 25.00 mg/mL, respectively). The HPLC–MS showed that USP rich in phenolic compounds, especially proanthocyanidin polymers. Moreover, we speculated that the antibacterial and antioxidant activity might be due to proanthocyanidin polymers. Therefore, UMSE is an efficient method to obtain phenolic compounds of AN, and USP is a potential antibacterial agent and antioxidant. © 2021, The Author(s), under exclusive licence to Springer Science+Business Media, LLC, part of Springer Nature.

摘要:
本研究旨在对人激肽释放酶7 (KLK7)基因进行原核与真核表达,研究其功能,并为后续KLK7新型生物抑制剂的研制提供基础材料。采用PCR扩增KLK7基因,将其克隆至原核表达载体pGEX4T-1和真核表达载体pEGFP-C1,构建重组表达质粒pGEX4T-1-KLK7和pEGFP-C1-KLK7,分别转化Transetta (DE3)和DH5α感受态细胞,筛选阳性菌株,用IPTG诱导重组KLK7蛋白(rKLK7)表达,SDS-PAGE和Western blot分析表达情况。将pEGFP-C1-KLK7转染人角质形成细胞HaCaT,观察KLK7在细胞中的定位,检测KLK7基因过表达对角质化细胞增殖的影响。KLK7基因开放阅读框为762 bp,编码254个aa。原核系统表达的rKLK7分子量为53.5 kDa。EGFP-KLK7融合蛋白定位在细胞质。KLK7的过表达对HaCaT细胞的增殖有一定的抑制作用,但差异不显著(P>0.05)。DNA检测表明,KLK7过表达可促进HaCaT细胞的凋亡。KLK7在原核表达系统和真核细胞中均成功表达,KLK7蛋白过表达对HaCaT细胞的增殖有一定的抑制作用,而对细胞凋亡有一定的促进作用。

Experiment was conducted to study the effects of Mulberry Leaf (ML) powder on reproductive performance, serum and milk amino acid composition in sows. Fifty sows (D 85 at gestation) with parity 3 or 4 were randomly divided into 5 groups: C, M100, M200, M300 and M400, receiving 0, 100, 200, 300 and 400 g ML powder per sow per day. Blood and milk of sows at Days 1 and 21 of lactation were collected. Results showed that average daily feed intake (ADFI) during lactation was higher in groups supplemented ML compared with control group (p < 0.01). Litter weight gain during lactation was higher in M400 than in groups M200 and C (p < 0.05), with no significant difference compared with M100 and M300. Serum glucose concentration in groups M400 and M300 was higher than those in the other groups (p < 0.01). Serum HDL-C concentration in group M400 was significantly greater than those in groups M100 and M200 (p < 0.05), with no significant difference between group M400 and groups M300, control. Milk amino acid concentrations such as isoleucine, leucine, lysine and valine were all lower in group M400 than in control (p < 0.01). Serum methionine (Met) concentration was higher in M300 than in other groups (p < 0.01). Milk Met concentration in group C was higher than those of the sows in the group M400, with no significant difference compared with groups M100, M200 and M300 (p < 0.05). Serum Lys and Met concentrations were lower in M400 than in control group (p < 0.05). In summary, our results have revealed the ML supplementation at a high dose such as 300 g/day during later gestation and lactation showed benefit in regulating lipid and amino acid metabolism in sows and then improved growth performance of their offspring.

摘要:
Peroxynitrite (ONOO−) is a highly reactive and short-lived oxidant with low content in cells, and plays an important role in biology. Accordingly, developing a rapid, sensitive, and effective detection approach of cellular ONOO− is very meaningful to further study its biological functions. In this work, a new biosensor BQ for the recognition of ONOO− was facilely synthesized by condensation of 2-hydrazinoquinoline and formylated benzothiazole possessing ‘‘ESIPT + AIE” feature. The probe can be used in a wide range of pH value, and displayed a rapid fluorescence “turn on” response toward ONOO− with low detection limit (5.1 nM) and high selectivity over other competitive substances through ONOO−-triggered oxidative cleavage of C=N double bond. The sensing mechanism of BQ towards ONOO− was proved by 1HNMR and HRMS analysis. Furthermore, the capacity of BQ to detect intracellular ONOO− has been well-demonstrated by fluorescence microscopic imaging experiments, indicating its potential application for investigating the physiological function of ONOO−. © 2022 Elsevier B.V.

摘要:
A tumor’s heterogeneity has important implications in terms of its clonal origin, progression, stemness, and drug resistance. Therefore, because of its significance in treatment, it is important to understand the gene expression pattern of a single cell, track gene expression or mutation in heterogeneous cells, evaluate the clonal origin of cancer cells, and determine the selective evolution of different subpopulations of cancer cells. Researchers are able to trace a cell’s mutation and identify different types of tumor cells by measuring the whole transcriptome with single-cell sequencing (scRNA-seq). This technology provides a better understanding of the molecular mechanisms driving tumor growth than that offered by traditional RNA sequencing methods. In addition, it has revealed changes in the mutations and functions of somatic cells as a tumor evolves; it has also clarified immune cell infiltration and activation. Research on scRNA-seq technology has recently advanced significantly, suggesting new strategies for the treatment of cancer. In short, cancer researchers have become increasingly dependent on scRNA-seq. This paper reviews the development, detection principles, and processes of scRNA-seq technology and their application in tumor research. 摘要:
In order to determine the effect of different oligosaccharides on growth performance, intestinal health, and antioxidant status of broilers, 240 1-day-old XiangHuang broilers were randomly distributed to 4 treatments with 6 replicates each. Birds were fed corn-soybean-based diets (CON), and birds in xylo-oligosaccharides (XOS), fructo-oligosaccharides (FOS), and iso-maltooligosaccharide (IMO) groups were given the basal diet supplemented with 200 mg/kg XOS, FOS, and IMO, respectively. Result showed that average daily gain (ADG) during the whole 5 weeks in FOS group was greater than that in control group (p < 0.05). Both breast and thigh muscle percentages were higher for birds fed XOS versus CON (p < 0.05). Oligosaccharides supplementation increased jejunal villus height compared with control group (p < 0.05). Malondialdehyde (MDA) concentration in breast muscle was lower for birds fed diet containing FOS versus CON (p < 0.05). Activities of total superoxide dismutase (T-SOD) in serum and thigh muscle were higher in IMO than in control group (p < 0.05). Serum T-SOD and breast muscle's glutathione peroxidase (GSH-Px) activity was higher in XOS compared with control group (p < 0.05). Conclusion, dietary oligosaccharides such as XOS, FOS, and IMO could improve intestinal health and antioxidant ability of muscle without affect growth performance in broilers.

The study investigated the effect of alfalfa hay substituted with ramie silage on the expression of apoptotic genes in the gastrointestinal tract of goats. Thirty-two goats were randomly allocated into four groups, in which the alfalfa was substituted with ramie at 0%, 35%, 75%, and 100% levels, respectively. In the rumen, the mRNA expression of Bax was significantly up-regulated (p =.0007) when alfalfa was 100% substituted by ramie; the mRNA expression of Bcl-2/Bax was significantly down-regulated (p =.02) when alfalfa was 100% substituted by ramie compared with the 75% substituted treatment; the protein expression of Bcl-xl was significantly down-regulated (p =.03) when alfalfa was 100% substituted by ramie compared with 35% and 75% substituted treatments, respectively. In the jejunum, the mRNA expression of p53 was significantly up-regulated (p =.01) when alfalfa was 100% substituted by ramie compared with 0% and 35% substituted treatments; the protein expression of p53 was significantly up-regulated (p =.001) when alfalfa was 35% substituted by ramie compared with 0% and 75% substituted treatments. However, the activity of Caspase-3 was not affected by different substituting levels of ramie in the rumen and jejunum of goats (p >.05). In conclusion, ramie with high substitution had strong antinutritional effect, which might promote the apoptosis in the gastrointestinal tract of goats in a caspase-independent manner, thus affecting the growth and development of goat. It was suggested that ramie should not replace alfalfa more than 35% in the process of goat feeding. © 2022 The Authors. Food Science & Nutrition published by Wiley Periodicals LLC.

ETHNOPHARMACOLOGICAL RELEVANCE: Salvia miltiorrhiza Bunge is a bulk medicinal material used in traditional Chinese medicine, that can cure cardiovascular diseases, neurasthenia, and other conditions. Sweating is a frequently used method of processing S. miltiorrhiza for medical applications. We previously demonstrated changes to the metabolic profile of linoleic acid, glyoxylate, and dicarboxylate after Sweating. However, this alteration has not been explained at the molecular level. MATERIALS AND METHODS: Fresh roots of Salvia miltiorrhiza Bunge were treated by the Sweating processing, and then the tandem mass tag technique was used to compare the proteome difference between Sweating S. miltiorrhiza and non-Sweating S. miltiorrhiza. RESULTS: We identified a total of 850 differentially expressed proteins after Sweating treatment in S. miltiorrhiza, including 529 upregulated proteins and 321 downregulated proteins. GO enrichment analysis indicated that these differentially expressed proteins are involved in external encapsulating structure, cell wall, oxidoreductase activity, ligase activity, and others. Further analysis showed that CYP450, the pathogenesis-related protein Bet v 1 family, and the peroxidase domain were the major protein domains. KEGG enrichment identified 18 pathways, of which phenylpropanoid biosynthesis is the most important one related to the metabolite profile and is the principal chemical component of S. miltiorrhiza. CONCLUSION: This study addressed potential molecular mechanisms in S. miltiorrhiza after Sweating, and the findings provide reasons for the changes in biochemical properties and metabolites changes which might cause pharmacological variation at the proteome level.
