摘要:
Simultaneous ultrasonic/microwave assisted hydrodistillation extraction (UMHE) was first employed to extract essential oil. Meanwhile, the effects of UMHE, hydrodistillation extraction (HE), microwave assisted hydrodistillation extraction (MHE) and solvent extraction (SE) on the yield, chemical composition, antibacterial and antibiofilm activity of essential oil were evaluated. The optimized conditions determined by response surface methodology were as follows: solvent to solid ratio was 5.2:1 (ml/g), extraction time was 15.5 min, and microwave power was 816 W. Moreover, 15 min was sufficient for UMHE to obtain higher yield than that by HE for 1.5 h, indicating a significant reduction of extraction time and an improvement of efficiency. Forty five compounds were identified in the essential oil through GC-MS analysis. The composition of essential oils obtained by UMHE, MHE and HE was similar, and the major compounds were D-Limonene, γ-Terpinene and α-Pinene. Moreover, the results indicated that the antibacterial activity of the essential oils was similar. All the essential oils exhibited inhibitive effect on biofilm formation. The inhibitive effect of essential oils of HE and UMHE was significantly higher than that of the essential oil obtained by SE. This study suggests that UMHE is a rapid and efficient alternative for the extraction of essential oils, with a nice potential for application. Also, UMHE provides a possible approach to improve the antibacterial and antibiofilm activity of essential oils. © 2018 Elsevier Ltd

摘要:
The name Hymenasplenium laterepens was first proposed by Cheng and Murakami in 1998 but was not validly published. We recently collected and studied this taxon at its locus classicus in Xishuangbanna, Yunnan Province in China. Molecular phylogenetic analyses and morphological observation of H. laterepens and related species clearly indicate that this is a distinct taxonomic entity worthy of species rank. Phylogenetically, H. laterepens is closely related to H. apogamum and may be involved in the reticulate evolution of the H. apogamum complex, which is comprised of several taxa with different levels of ploidy. Morphologically, H. laterepens is distinguished by the combination of its triangular laminae, large pinnae with acute apices and deeply serrated margins, and ca. 117 chromosomes at meiosis I and 32 spores in each sporangium. This work provides a complete species description and comparison to related species within the genus Hymenasplenium, particularly to other relatives of the “H. unilaterale” group. Hymenasplenium laterepens is described here with diagnosis and type designation. © 2018 Magnolia Press.

摘要:
以模拟含铜废水为研究对象,利用响应曲面法对D401螯合树脂去除废水中铜的操作条件进行优化。优化了单因素的影响,并在此基础上选取pH值、温度、Cu~(2+)的初始质量浓度作为自变量,以吸附量为响应值,根据Box-Behnken实验设计原理,采用三因素三水平响应曲面设计对吸附量进行回归分析,模拟得到二次多项式回归方程的预测模型。根据响应曲面图优化结果和实际操作条件,确定最佳条件为:pH值4.35,温度40.00℃,Cu~(2+)的初始质量浓度620mg/L,最大吸附量83.34mg/g,与模型理论预测值82.64mg/g基本吻合。结果表明,模拟得到的预测模型可靠,响应曲面法可以用于含铜废水中铜的去除特性的优化。

摘要:
α-酮戊二酸(AKG)是三羧酸循环中的中心物质,也是氨基酸和碳水化合物代谢的桥梁。AKG作为铵离子清除剂,是提供谷氨酰胺的来源,能够促进肌肉蛋白质合成和抑制蛋白质降解,并为胃肠道细胞提供能量代谢的燃料。AKG可以通过脯氨酰-4-羟化酶产生脯氨酸,增加胶原蛋白合成,促进骨骼系统发育。此外,AKG可以抑制ATP合成酶和雷帕霉素靶蛋白,延长线虫的寿命。AKG不仅延长寿命,而且延迟与年龄相关的疾病。另外,AKG在消除毒性、提高耐寒性、缓解氧化应激损伤以及促进人类多潜能干细胞分化等方面具有关键作用。本文旨在对AKG的生理功能及其在动物生产领域的应用进行综述,以促进人们对AKG的理解,为AKG在动物营养调控和生产实践中的应用提供理论依据。

摘要:
运用实验室分子诊断技术对湖南岳阳某猪场一例疑似猪繁殖与呼吸综合征病毒(PRRSV)感染病例进行确诊,测定并分析其基因序列,为猪蓝耳病的防控提供依据。采集病猪淋巴组织提取RNA,采用逆转录—聚合酶链式反应(RT-PCR)检测病毒核酸,构建重组载体,经序列测定,利用DNAStar等生物信息学软件进行序列分析。结果显示,R T-PCR产物大小为657 bp,同源性比较结果显示,此毒株OR F5基因核苷酸序列与与经典美洲型毒株VR-2332的同源性为83.4%,与我国经典毒株CH-1a株的同源性为85.4%,与高致病性代表株HUN4和JXA1的同源性分别为84.5%和84.3%,与欧洲经典型毒株LV的同源性为62.8%,与GDZS2016和GD/YJ/2014的同源性分别为95.0%和95.9%。遗传进化分析表明,此毒株属美洲型毒株;美洲型毒株下分两个亚群,以近年来新流行的GDZS2016和GD/YJ/2014毒株为代表的Ⅰ亚群和以HuN4、JXA1和VR-2332等为代表的Ⅱ亚群,此毒株与GDZS2016和GD/YJ/2014等同属Ⅰ亚群。结果表明,此次病例是由美洲型PR R SV毒株感染引起,将此毒株命名为HuNYY201805。

摘要:
对一例疑似犬瘟热(CD)的病例进行诊断及治疗,分别采用胶体金试纸条检测、采用逆转录-聚合酶链式反应(R T-PCR)检测病原抗原和犬瘟热病毒(CDV)N基因序列分析。结果显示,病犬鼻分泌物经胶体金检测为CDV抗原弱阳性,RT-PCR产物大小为287 bp,此毒株N基因核苷酸序列与野毒株的核苷酸序列同源性高达96.5%~98.6%,遗传进化分析显示,该毒株与疫苗株(Onderstepoort、CDV3等)和Lederle等弱毒株不在同一分支上,而与野毒株在同一分支,说明该毒株为CDV强毒株,将此分离株命名为CDV-Hu NHY171124。采用中西结合的抗病毒治疗方案对病犬进行为期7 d的治疗,疗效不显著,病情恶化而接受安乐死。

摘要:
为确诊衡阳市某猪场育肥猪发生的疑似猪伪狂犬病(PR)疫情,采集发病猪脾脏,运用PCR方法进行猪伪狂犬病病毒(PRV)核酸检测,利用非洲绿猴肾细胞(Vero)进行病毒分离培养,采用免疫过氧化物酶单层细胞染色法(IPMA)进行病毒血清学鉴定;用分离培养物进行家兔感染试验,观察临床症状和病例剖检变化,采用PCR检测各器官中的病毒分布情况.结果显示:病料接种Vero细胞培养3 d后,出现明显的细胞病变效应(CPE);分离培养的病毒与PRV阳性血清呈阳性反应;家兔感染后出现奇痒等神经症状并死亡,其肝、脾组织均为PRV gE核酸检测阳性.试验结果证实,分离到的病毒为PRV野毒,从而确诊了该疑似疫情.

以罗汉果粉末为原料,乙醇为提取溶剂,采用超声波辅助法提取罗汉果糖甙.通过单因素和正交试验探究超声波时间、乙醇浓度、料液比对罗汉果糖甙提取的影响.结果表明:最佳提取条件是95%乙醇、料液比1:20、超声波时间40 min,在此条件下罗汉果糖甙的提取含量为694μg.

摘要:
以滤纸条为唯一碳源培养基对富含纤维素的土样微生物进行富集培养,再用刚果红纤维素琼脂培养基筛选降解效果好的菌株,根据菌落形态及显微结构进行鉴定,并测定其发酵液的FPAase酶活和对稻草秸秆的降解情况。通过刚果红纤维素琼脂培养基筛选得到的最具纤维素分解能力的真菌经形态鉴定为冬克青霉(Penicillium donkii),该菌在含有羧甲基纤维素钠的液体培养基中28℃,180 r/min培养,在第8d酶活力达到最大为15.0 U/m L。接种菌株21d的稻草纤维素含量由接种前的30.30%减少至26.36%。青霉1-4为冬克青霉,对纤维素有一定的降解能力,可以加速对稻草秸秆中纤维素的降解。

摘要:
The floral organogenesis and anatomy of Koelreuteria bipinnata and its variety K. bipinnata var. integrifolia (Sapindaceae) has been investigated to clarify the identity of the two taxa in relation to other species of Koelreuteria, and to understand the shift to monosymmetry in the genus. Although the floral development is highly similar, we found a number of striking differences. Flowers arise in thyrses, with lateral branches forming cincinni of 5–9 flowers. Sepals initiate in a spiral sequence. Five petals arise unidirectionally alternating to the sepals. The last formed petal and one stamen between sepals 3 and 5 are strongly delayed, appearing as a common primordium, while this petal is completely suppressed in var. integrifoliola. Eight stamens initiate sequentially, differ in size and partly precede the development of petals. The gynoecium develops as a triangular primordium on which three carpellary lobes become demarcated simultaneously. Placentation is axile. Septal slits occurring within the style are interpreted as a deep-reaching non-nectariferous extension of the stigma. The massive, oblique disk with crenate apex develops in an extrastaminal position, but is interrupted on the radius of the lost petal. Floral developmental evidence supports variety K. bipinnata var. integrifolia rather than being synonym of K. bipinnata. Floral development is compared with K. paniculata and is discussed in the context of floral evolution of Sapindaceae. Our study demonstrates the importance of developmental shifts on floral evolution. The triangular gynoecium has a strong spatial impact in obliquely reorganizing the symmetry of the flower. It is demonstrated that spatial constraints of calyx and ovary are responsible for the reduction in one of the petals, two stamens and a shift in symmetry of the flower. © 2018, Springer-Verlag GmbH Austria, part of Springer Nature.

以南瓜为原料,以花生油、卡拉胶和木糖醇为辅料,研制低热量南瓜月饼馅料.通过单因素试验探讨花生油添加量、卡拉胶添加量、木糖醇添加量以及炒制时间对馅料感官品质的影响,从而确定各因素的最佳水平,进而利用正交试验法确定南瓜月饼馅料的最优加工工艺为:以南瓜泥质量为100%计,花生油添加量5%、卡拉胶添加量0.6%,木糖醇添加量3%,炒制时间30 min.所得月饼馅料甜度适当、口感细腻、具有明显的南瓜风味.

摘要:
【目的】制备猪流行性腹泻病毒(PEDV)CH-HeB14毒株S1蛋白的多克隆抗体,为PEDV新流行毒株诊断试剂盒和疫苗研制提供基础材料。【方法】利用RT-PCR扩增PEDV CH-HeB14毒株S1基因,对序列特征进行生物信息学分析,用PCR将S1基因截成不同片段(S1A、S1B、S1C、S1C-1和S1C-2),分别克隆至原核表达载体pET28a,转化BL21(DE3)构建重组表达菌株;用IPTG诱导重组蛋白表达,并进行SDS-PAGE和Western blotting鉴定。将表达蛋白与佐剂乳化制备免疫原免疫兔多克隆抗体,Western blotting和免疫过氧化物酶单层细胞染色法(IPMA)检测抗体的免疫活性。【结果】 PEDV S1、S1A、S1B、S1C、S1C-1和S1C-2片段长度依次为2 079,600,600,878,549和447 bp。系统进化树表明,CH-HeB14株与2010年以来的22个新流行毒株亲缘关系较近,而与经典疫苗株CV777、attenuated DR13亲缘关系较远。成功截短表达了S1蛋白不同区段S1A、S1B和S1C-1重组蛋白(rS1A、rS1B和rS1C-1),且蛋白均以包涵体形式表达。制备了抗rS1A、rS1B和rS1C-1蛋白的多克隆抗体,其与这3种蛋白反应良好,且抗rS1A重组蛋白的多克隆抗体Western blotting效价为1∶32 000;IPMA结果显示,该抗体能与PEDV CH-HeB14毒株和经典疫苗毒株attenuated DR13均发生特异性反应。【结论】成功克隆、表达了变异毒株PEDV CH-HeB14的S1基因,所制备的S1蛋白多克隆抗体具有良好的免疫活性。

摘要:
To provide information on reclamation of multi-heavy metal polluted soils with conception of phytostabilization, a field survey on the uptake and accumulation of potentially toxic elements such as antimony (Sb), arsenic (As), lead (Pb), cadmium (Cd), copper (Cu), and zinc (Zn) in colonized plant species around the world’s largest antimony mine area, China, was conducted. Samples including leaves and shoots (including roots and stems) of colonized plants as well as rhizospheric soils were collected from eight sampling zones in the studied area. The results showed that the contents of Cu, Zn, and Pb in rhizospheric soils below plants were comparable to the corresponding background values of Hunan province, otherwise Sb, Cd, and As contents were extremely high (17–106, 17–87, and 3–7 times of the corresponding background values). The highest concentration of Sb was found in Aster subulatus (410 mg kg −1 ); Cd, As, and Zn were in Herba bidentis bipinnatae (10.9, 264, and 265 mg kg −1 , respectively); and Cu was in Artemisia lavandulaefolia (27.1 mg kg −1 ). It also exhibited that all the contents of As in leaves were several times of those in shoots of plants, Cd and other heavy metals showed in a similar pattern in several studied species, implying that the uptake route of these heavy metals via foliar might contribute to the accumulation. With high bioconcentration factors of heavy metals (more than 1, except for Zn), together with the growth abundance, Herba bidentis bipinnatae was considered as the most suitable colonized species for phytostabilization of the multi-heavy metal pollution in soils on this antimony mine area. © 2018, Springer Science+Business Media B.V., part of Springer Nature.

1886年Franchet描述并发表茶条木Delavaya toxocarpa,他引用了Delavay所采集的标本(J. M. Delavayn. 903).我们对原始文献考证发现,这些标本包括了采于不同时期的花期及果期材料,包括了两份以上的标本,但作者在发表该名称时没有明确指定其中的一份标本作为模式标本,因此,很有必要对其进行后选模式标本指定.为此,我们根据墨尔本国际植物学命名法规9.2和9.11,为其指定了后选模式标本.

摘要:
为了改善柑橘皮的营养价值,试验以粗蛋白提高幅度为衡量标准,采用单因素试验设计探讨黑曲霉发酵柑橘皮的最适宜发酵时间、接种比例、底物组合以及水的添加量。结果表明:当底物红薯与柑橘皮添加比例为1∶1,加水10 mL,接种10%的黑曲霉孢子悬液,发酵第4天时发酵产物的粗蛋白含量最高(8.23%),较发酵前提高了41.17%;发酵产物的粗脂肪含量从1.71%提高到2.13%,提高了24.56%。说明通过黑曲霉固态发酵可以增加柑橘皮的粗蛋白含量。

Campylobacter is a well-known food-borne pathogen that causes human gastroenteritis. Food products that contain Campylobacter may also be contaminated by other pathogens, however, whether this multiple contamination leads to more severe infection remains unclear. In this study, mice were gavaged with Campylobacter jejuni and other food-borne pathogenic bacteria to mimic a multiple infection. It was demonstrated that the C. jejuni load was elevated when the mice were co-infected with C. jejuni and Salmonella typhimurium, and the campylobacteriosis that followed was also enhanced, with features of decreased body weight, heavier bloody stools and more pronounced inflammatory changes to the colon. In addition, infection with C. jejuni was also promoted by co-infection with entero-invasive Escherichia coli but unaffected over time. In contrast to S. typhimurium and entero-invasive E. coli, co-infection by Listeria monocytogenes showed little effect on C. jejuni infection and even hindered its progress. In addition, the intestinal microecology was also affected by co-infection of C. jejuni with other pathogens, with an increased relative abundance of unclassified Enterobacteriaceae, decreased levels of butyric acid and changes in the abundance of several genera of gut microbe, which suggests that some food-borne pathogenic bacteria might affect the progression of C. jejuni infection in mice by influencing the composition of the gut microbiota and the resulting changes in SCFA levels. Collectively, our findings suggest that co-infection of Campylobacter with other pathogenic bacteria can impact on the progression of infection by C. jejuni in mice, which may also have implication for the etiology of Campylobacter on human health. © 2018 Wang, He, Jin, Zhou, Chen, Zhao, Zhang and Chen.

Behavioral and Electrophysiological Responses to Caffeine of Helicoverpa armigera Larvae and H. assulta Larvae

The dual-choices tests of behavioral test were used to study the gustatory behavioral responses to caffeine of Helicoverpa armigera larvae and H. assulta larvae. Electrophysiological responses were studied by electrophysiological tip recording. Behavioral experiments showed that caffeine was a feeding deterrent for both larvae,but it showed a stronger feeding deterring effect on the oligophagous H. assulta. The electrophysiological tip-recording results showed that both H. armigera larvae and H. assulta larvae had one caffeine-sensitive feeding deterrent neuron at the medial sensilla,and the electrophysiological pulse response of H. assulta larvae was significantly stronger than that of H. armigera larvae. Therefore,caffeine had a stronger feeding deterring effect on the oligophagous H. assulta than the polyphagous H. armigera. The difference in behavioral effect was closely related to the sensitivity of to caffeine the feeding deterrent neurons at the medial sensilla.
