HACCP原理通过危害分析确定关键控制点、建立关键点监控体系与纠正措施,将质量安全管理从源头控制。借鉴SWOT-HACCP原理、利用相关的教育学理论,对环境工程专业课堂教学过程的关键控制点进行分析,建立监控与纠错体系,有利于指导老师开展教学活动,保证学生培养质量从源头控制,对环境工程专业课程教学管理具有十分重要的指导意义。

摘要:
Salvia miltiorrhiza has numerous compounds with extensive clinical application. “Sweating”, a processing method of Traditional Chinese Medicine (TCM), results in great changes in pharmacology and pharmacodynamics. Previously, chromatogram of 10 characteristic metabolites in S. miltiorrhiza showed a significant difference after “Sweating”. Due to the complexity of TCM, changes in metabolites should be investigated metabolome-wide after “Sweating”. An untargeted UPLC/MS-based metabolomics was performed to discover metabolites profile variation of S. miltiorrhiza after “Sweating”. Multivariate analysis was conducted to screen differential metabolites. Analysis indicated distinct differences between sweated and non-sweated samples. 10,108 substance peaks had been detected altogether, and 4759 metabolites had been identified from negative and positive ion model. 287 differential metabolites were screened including 112 up-regulated and 175 down-regulated and they belong to lipids and lipid-like molecules, and phenylpropanoid and polyketides. KEGG analysis showed the pathway of linoleic acid metabolism, and glyoxylate and dicarboxylate metabolism were mainly enriched. 31 and 49 identified metabolites were exclusively detected in SSM and NSSM, respectively, which mainly belong to carboxylic acids and derivatives, polyketides and fatty acyls. By mapping tanshinones and salvianolic acids to 4759 identified metabolites library, 23 characteristic metabolites had been identified, among which 11 metabolites changed most. We conclude that “Sweating’’ has significant effect on metabolites content and composition of S. miltiorrhiza. © 2020, The Author(s).

摘要:
该实验旨在通过对发酵条件进行优化,提高链霉菌PTY-3绿色素的产量。以分离自吉林省四平市大棚黑土中的一株链霉菌PTY-3为实验菌株,对发酵时间、装液量、接种量、发酵起始pH和发酵温度进行了优化。结果表明,在发酵时间8 d,发酵温度为28℃,接种量为10%,装液量110 m L/250 mL和发酵起始pH值为7.5的条件下培养时绿色素的色价最高,可达77.0 U/m L,是未优化前(57.3 U/mL)的1.34倍。该研究为天然绿色素的进一步工业化生产提供了科学依据。

摘要:
为了生产优质速冻糄粑,通过复水温度、复水时间、熟制方式、速冻时间和解冻时间预实验,选取糄粑的预冷温度、预冷时间、速冻温度等三个因素进行正交实验,通过对速冻糄粑熟制后的感官评定、硬度、粘聚性、弹性、内聚性、胶粘性、咀嚼性、回复力等为参数进行品质评价。实验结果表明:糄粑出窝后直接复水比干制后复水更加适用于生产,蒸制方式更适应市场的发展,速冻后直接熟制比解冻1 h后熟制更加节约时间,确定复水水温40℃、复水时间20 min、预冷温度0℃、预冷时间40 min、速冻温度-40℃、速冻时间40 min,此工艺得出速冻糄粑具有最优的感官品质和质构,有利于提高速冻糄粑的市场竞争力。

摘要:
豆腐乳是我国传统特色美食,也是乳酸菌的天然菌库。文章以寻求适宜人体的益生性乳酸菌为目标,采用溶钙圈法从南岳茶油豆腐乳中分离出6株乳酸菌,编号为RSP1~6。结果表明:6株菌均为球状乳酸菌,RSP5和RSP6可在pH 4.0、 11.0%NaCl、 0.35%胆盐条件下存活,RSP5的耐酸、耐盐能力最强;RSP6的耐胆盐、产酸能力最强,且生长活性最强、生长周期最短。将RSP5和RSP6菌株进行16S rDNA测序并构建系统发育树,发现RSP5、RSP6与戊糖片球菌亲缘最近,鉴定为戊糖片球菌。

摘要:
考察了唾液乳杆菌(Lactobacillus salivarius)CCFM 1054体外培养产酸及发酵上清液抑制空肠弯曲杆菌(Campylobacter jejuni)生长能力、对人工模拟胃肠液中的耐受、对共培养条件下的抑菌能力、对HT-29细胞的粘附以及自我形成生物膜的能力,并以鼠李糖乳杆菌LGG和植物乳杆菌N49作为对比菌株,分别干预空肠弯曲杆菌和弓形虫复合感染的小鼠。结果显示,CCFM 1054能显著改变小鼠肠道菌群的组成,降低空肠弯曲杆菌在小鼠体内的定植率并缓解其感染。肠道菌群变化和乳酸菌拮抗空肠弯曲杆菌相关的体内外特性的相关性分析表明,CCFM 1054对细胞的高粘附性及其较强的生物膜形成能力使得其能在小鼠体内显著改变肠道菌群丰度。

摘要:
The venom of Lycosoidea spiders is a complex multicomponent mixture of neurotoxic peptides (main components) and antimicrobial peptides (AMPs) as minor components. In this study, we described the high-throughput identification and analysis of AMPs from Lycosa sinensis venom (named LS-AMPs) using a combination strategy that includes the following three different analysis approaches: (i) peptidomic analysis, namely reversed-phase high-performance liquid chromatography (RP-HPLC) separation plus top-down sequencing by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MS); (ii) transcriptomic analysis, namely cDNA library construction plus DNA sequencing; (iii) bioinformatic analysis, namely analysis and prediction for molecular characters of LS-AMPs by the online biology databases. In total, 52 sequences of AMPs were identified from L. sinensis venom, and all AMPs can be categorized into eight different families according to phylogenetic analysis and sequence identity. This is the largest number of AMPs identified from a spider species so far. In the present study, we demonstrated molecular characteristics, such as complex precursor, N- and/or C-terminally truncated analogs, and C-terminal amidation of LS-AMPs from L. sinensis venom. This is a preliminary investigation on the molecular diversification of venom-derived AMPs from the wolf spider species (family Lycosidae), and a detailed investigation on the functional diversity of LS-AMPs will be preformed in the future.

摘要:
豆花是一种人们喜爱的大豆加工食品,一般采用盐卤或石膏作为凝固剂制作,但卤水点的豆花产量较低,还可能产生苦涩味。本文以黄豆为原材料,以葡萄糖酸-δ-内酯(GDL)为凝固剂,利用湿法工艺制备内酯豆花。在单因素试验基础上,选取料水比、凝固温度和内酯添加量为影响因素,以内酯豆花的凝胶强度和感官评分作为响应值,开展实验,优化内酯豆花制作工艺。结果表明,较佳工艺条件为:料水比1∶15(g/mL),凝固温度为90℃,添加0.20%内酯,凝固20min。此工艺条件制备的豆花凝胶状态好、口感滑嫩、色泽均一、出水少,潜在市场价值巨大。

摘要:
Camellia oleifera shell-based activated carbon (COSAC) was prepared by H3PO4 activation method and further used to remove U(VI) from the aqueous solution in a batch system. This research examined the influence of various factors affecting U(VI) removal, including contact time, pH, initial U(VI) concentration, and temperature. The results showed that the U(VI) adsorption capacity and removal efficiency reached 71.28 mg/g and 89.1% at the initial U(VI) concentration of 160 mg/L, temperature of 298 K, pH 5.5, contact time of 60 min, and COSAC dosage of 2.0 g/L. The pseudo-first-order, pseudo-second-order, and intraparticle diffusion equations were used to identify the optimum model that can describe the U(VI) adsorption kinetics. The pseudo-second-order kinetics model performed better in characterizing the adsorption system compared with the pseudo-first-order and intraparticle diffusion models. Isotherm data were also discussed with regard to the appropriacy of Langmuir, Freundlich, Temkin, and Dubinin-Radushkevich models. The Langmuir model described the U(VI) adsorption process the best with a maximum adsorption capacity of 78.93 mg/g. Thermodynamic analysis (Delta G(0) < 0, Delta H-0 > 0, and Delta S-0 > 0) indicated that the U(VI) adsorption process is endothermic and spontaneous. All the results imply that COSAC has a promising application in the removal or recovery of U(VI) from aqueous solutions.

目的:利用GC-MS技术研究不同配方发酵糙米乳的挥发性风味物质组成,并结合感官评定指标评估发酵糙米乳的产品品质,以期对发芽糙米乳新产品的开发及感官品质提升,提供理论依据。方法:选用不同配方的发芽糙米乳,采用气相色谱-质谱联用技术对发芽糙米乳产品的挥发性风味物质进行测定,经偏最小二乘回归法对感官属性与挥发性风味物质成分做相关性分析。结果:经GC-MS分析并通过数据库检索结合质谱信息比对,筛选得到21种挥发性风味物质。结论:2,3-丁二酮、乙偶姻、2,3-戊二酮、1-庚醇和乙酸分别与奶香味、米香味和酸味呈显著相关,是发芽糙米乳的主要风味物质。

Main findings: The Sb resistant bacterium impacted the fraction and quantity of IP on rice roots, and it decreased the Sb concentrations in rice shoots. © 2019 Elsevier LtdIron plaque (IP) is crucial in mitigating antimony (Sb) uptake and accumulation in rice plants, while, few studies focused on the effect of the iron plaque-associated Sb resistant bacteria on IP and Sb uptake into rice plants. Here, the effect of a Sb resistant bacterium (GenBank accession No. MH345840, with potential of conversion soluble Sb(III) into insoluble Sb2O3) on IP and Sb(III)/Sb(V) uptake under hydroponic condition was investigated. The results showed that in the presence of Sb(III), a large quantity of bacterial cells consorted with IP on rice roots, the bacterial inoculum altered the IP fraction distribution without enhancing its amount. However, it reduced Sb(III) uptake into rice roots. On contrary, seldom bacterial cells associated with the IP on rice roots in the presence of the Sb(V), the bacterial inoculum increased the IP amount slightly, and did not decline the Sb(V) uptake into rice roots. It also showed that the bacterial inoculum decreased Sb concentrations in rice shoots greatly in both Sb(III) and Sb(V) supplied treatments. © 2019 Elsevier Ltd

摘要:
研究旨在克隆猪流行性腹泻病毒(PEDV)HuN-HY1902毒株N、sM、ORF3基因并进行序列分析,为PEDV的临床防控提供依据。采用Vero细胞进行病毒分离培养,免疫过氧化酶单层细胞染色法进行病毒鉴定;RT-PCR扩增PEDV HuN1902毒株N、S、sM、ORF3基因,经纯化后进行序列测定,利用DNAStar、Mega等生物信息学软件进行序列分析。R T-PCR扩增得到PEDV ORF3、N、sM片段大小依次为744 bp、1400 bp、和1100 bp。该毒株ORF3基因核苷酸序列与PEDV/USA/Missouri130/2015毒株相似性最高,为99.7%;sM基因核苷酸序列与CH/SCMY/2018和BJ-2011-1相似性最高毒株,为96.3%;N基因核苷酸序列与CH/SCMY/2018毒株相似性最高,为98.6%;ORF3、N和sM基因核苷酸序列与疫苗毒株CV777依次为96%、95.1%和93.9%。进化分析显示,该毒株的ORF3、sM和N基因的核苷酸序列与2018年流行毒株CH/SCMY/2018处于同一分支,而与经典毒株CV777、SM98的亲缘关系较远。所分离培养的毒株命名为PEDV HuN-HY1902毒株,为新流行的变异毒株。

摘要:
Leaf architecture of 30 species representing 2 genera of Aceraceae in China was investigated for the first time. Within the 30 species, 10 species are found with an actinodromous venation pattern, while 20 species are found with a pinnate venation pattern. Within the 20 species with pinnate venation, 10 species are camptodromous, while the other 10 species are craspedodromous. Four types of leaf teeth are detected, viz. entire, toothed, lobed and partite. The secondary veins of most species are unbranched, except for Acer cordatum. Most species have obvious intersecondary veins, except for 4 species, i.e., Dipteronia sinensis, D. dyerana, Acer davidii and A. maximowiczii. All species have both reticulate and percurrent tertiary veins, except for A. pentaphyllum, which has only reticulate tertiary veins. All species are found with irregular areoles, except for A. pentaphyllum which is found to be regular. Veinlets are simple, branched or absent. A key to the genera and sections ofAceraceae mainly based on leaf architecture characters is presented.

Floral Development Reveals the Existence of a Fifth Staminode on the Labellum of Basal Globbeae

We observed the floral development of Hemiorchis burmanica and two species of Gagnepainia, which make up the sister group of Globba in Globbeae, as well as a selected number of species of Globba. Our observations revealed that in Gagnepainia, a “fifth staminode,” develops as part of the labellum, while it is generally lost in other Zingiberaceae. The fifth staminode, however, was not initiated in the labellum of Hemiorchis. A fifth staminode was also observed in Globba geoffrayi but was not found in all other species of Globba investigated. While the staminode becomes an integral part of the labellum in Gagnepainia, it remains small and almost aborts at later stages of development in G. geoffrayi. These results indicate that the development of a fifth staminode could be a recurring plesiomorphic trait for Globbeae, only retained in G. geoffrayi. The retention of the fifth staminode and its heterochronic shift may be linked with the mechanical constriction within the flower bud. 摘要:
The active sites of a mixed Cu/Ce material and the doping effect of typical element (iron, Fe) on the active species and the catalytic behavior of Cu/Ce for CO preferential oxidation in rich H 2 (CO-PROX) were investigated by in situ diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS), in situ oxygen storage capacity measurement (OSC) combined with designed temperature-programmed reduction (TPR), along with Raman, X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), and temperature-programmed desorption/reduction of CO (CO-TPD/TPR). These results showed that two kinds of surface active center were involved in the CuCe- and Fe-doped CuCe systems, that is, Cu + as adsorption sites for the chemisorption and the activation of CO molecules, the surface reactive oxygen (the highly dispersed oxygen and surface lattice oxygen) that directly participated in the whole CO oxidation process. The addition of Fe into CuCe sample resulted in the incorporation of Fe into CeO 2 lattice forming Fe-O-Ce structure and generated more oxygen vacancies, which not only enhanced the interaction between Cu and Ce to form more Cu + absorption sites but also trapped the gas-phase oxygen and promoted the release of subsurface lattice oxygen to supply more reactive oxygen. Thus, the turnover frequency (TOF) value was increased from 3.62 × 10 -2 s -1 for CuCe to 4.50 × 10 -2 s -1 for Fe-doped CuCe. Moreover, with the enhancement of the lattice oxygen migration combined with the promotional role of Fe on the water gas shift (WGS), the capacity of the resistance to CO 2 and H 2 O was enhanced for Fe-doping CuCe, and the corresponding stability time was largely prolonged from 170 to 400 h, in the coexistence of CO 2 and H 2 O. © 2019 American Chemical Society.

Spider venoms are insecticidal mixtures with diverse biological activities, and acylpolyamines are their small molecular active components. However, the mechanism for the insecticidal activity of acylpolyamines remains to be elucidated. Here, the structure and function of two acylpolyamine toxins, AVTX-622 and AVTX-636, from Araneus ventricosus were investigated. Nuclear magnetic resonance (NMR) analysis illustrated that the structure of two toxins was very similar, and compared to AVTX-636, AVTX-622 only missed a methylene group in the linker region between the polyamine head and tail. Both the two toxins did not inhibit on voltage-gated sodium channels in mammalian neuronal cells. Intriguingly, AVTX-622, but not AVTX-636, inhibited voltage-gated sodium channels in DUM neuronal cells of Periplaneta americana. Further animal test displayed that the paralyzing potency of AVTX-622 on insect was over ten-times stronger than that of AVTX-636. These findings indicate that a single methylene deletion from AVTX-636 offered AVTX-622 the insect-selective voltage-gated sodium channel activity, which not only elucidated structure-function of the toxins, but also provided new clues for insect-selective insecticide design. © 2019 American Chemical Society.

摘要:
为了使生菜种子能快速萌发节约生产成本和时间,将植物激素赤霉素作为加快其萌发生长的促进剂。本试验选取菊科莴苣属的罗马直立生菜(Lactuca sativa var. ramosa Hort)作为试验材料,设置不同的赤霉素浓度梯度(0. 001,0.01,0.1,1,10,100μmol·L-1),研究赤霉素对生菜种子的萌发及早期发育的影响。结果表明,与CK相比,赤霉素GA3有助于提高罗马直立生菜种子的发芽率、发芽势、发芽指数、活力指数及幼苗的根长、子叶长和鲜质量,各指标随着GA3浓度的升高均呈先升后降的趋势,分别于1,1,1,10,0.1,10,10和0.001μmol·L-1达到最大值,与CK差异显著;罗马直立生菜幼苗下胚轴长度随着GA3的升高而增加。综合各项指标,以1~10μmol·L-1浓度的GA3在促进罗马直立生菜种子萌发和早期发育方面效果最佳。

平菇下脚料对空心菜生长和产量的影响

摘要:
平菇下脚料富含氮磷钾等营养物质,可作为有机肥应用于蔬菜种植方面,根据平菇下脚料和土壤量设置6个处理,即0g+3 250 g(对照),50 g+3 200 g,100 g+3 150 g,150 g+3 100 g,200 g+3 050 g,250 g+3 000 g,采用盆栽试验研究不同处理空心菜株高、茎粗、主根长以及产量等指标的差异,探讨平菇下脚料在空心菜生产上的最佳施用量。结果表明,每盆施用50,100,150 g平菇下脚料均可在一定程度上提高空心菜株高、茎粗、根长及产量,其中以每盆100 g添加量效果最佳,即平菇下脚料与土壤的最佳配比为100∶3 150,空心菜产量较对照增加了40.9%;每盆施用200,250 g平菇下脚料的空心菜株高、茎粗、根长和产量均低于对照,说明平菇下脚料施用量过高对空心菜生长存在抑制效应。
